Was Thanos’ blade the strongest weapon in the MCU?
THIS ANSWER INCLUDES SPOILERS ABOUT ENDGAME. YOU'VE BEEN WARNED.

It might be.

The MCU has setup Vibranium to be the strongest known metal. But that's just it, known.

Hands down, Vibranium is the strongest metal on Earth, but in a universe as large as Marvel? Yeah, there's definitely metals out there much stronger.

It was shown in the first Avengers that the impact of Mjolnir charge with thunder did nothing to Captain America's shield. However, that was only one hit. What if Thor repeatedly hit Cap's shield? I'm pretty sure that eventually, it would suffer the same fate as it did Endgame.

Vibranium is strong, very strong, but not indestructible.

The most obvious evidence for this is in Endgame when Thanos kept hitting Cap's shield with his blade and it eventually broke, just like in Tony's vision. I guess being the brute Thanos is has something to do with it too.

Speaking about vision, Vision was also made entirely of Vibranium, but what happened to him? Died. Corvus Glaive's scythe went straight through pure Vibranium.

“But his scythe wasn't any ordinary scythe!”

Correct, but Vibranium isn't just any ordinary metal either.

Corvus Glaive was an adopted child of Thanos. If he has such a powerful weapon capable of penetrating Vibranium, then Thanos, the biggest and baddest villain the MCU has seen yet, will have a weapon equal to or greater than Corvus's scythe.

Did we just forget that Thanos plucked the mind stone straight out of Vision's head with his bare hands? If he can break Vibranium with his hands, then he can easily do it with the help of a weapon.

So yes, it seems that whatever Thanos's blade was made of, it is the strongest material we've seen so far in the MCU.

Is his sword the strongest weapon in the MCU? Now that can be debated.
