Absolutely. The class of bus you want is called executivo or de lujo (luxury). This is a step above first class. The two companies I’m most familiar with are ETN and Touristar. Lots of leg room, and the seats are wide. On the left side of the aisle is a single seat and on the right side it’s two seats. It’s more comfy than a lot of airplane cabins I’ve been in. Most of them have wifi. Two bathrooms in the back. Modern Mercedez buses. Oh, and you get a sack lunch when you board! There are stops at some of the bigger cities, especially towards the beginning of the trip, but not a lot. For example, taking the bus from Laredo, you’ll probably stop at Monterrey. Then either Matehuala or San Luis Potosí. (Incidentally, if you want to check out Real de Catorce, you reach it from Matehuala https://en.wikipedia.org/wiki/Real_de_Catorce ).

I’m living in San Miguel de Allende (https://en.wikipedia.org/wiki/San_Miguel_de_Allende), which is a big tourist destination. However, I highly recommend the city of Guanajuato (https://en.wikipedia.org/wiki/Guanajuato_City). From either town you can take a bus to Guadalajara and Puerta Vallarta or, of course, Mexico City and points south.
