How does a high resolution & high quality X-ray crystal structure of a protein target or protein-ligand complex help in drug discovery? By revealing very detailed information about amino acid side chain interactions and importantly, water-mediated interactions. An example from our very high resolution/ultra-high resolution 1.25A crystal structure of a protein, from crystals that we recently optimized for a partner. Previous structures of this protein (in the PDB and literature) were determined at 1.9A-2.1A, providing us with a significant jump in resolution. In the majority of our cases, we are able to improve crystal structure resolution than previously determined using a combination of crystal optimization and X-ray diffraction data collection & processing strategies. There is a lot that can be done on a target protein to improve crystallization and crystallography beyond production of crystallization-grade protein.
