Procedure

Sift 30g of soil to make one consistency.

Mix sample with equiv. 10g of anhydrous sodium sulfate placed in container, labeled, date and locations then left to dry for 24hrs

Extraction

1. Pour 50ml methychloride & 50ml of acetone into beaker with 30g of prepared dried soil sample

2. Place mixture in the Sonicator, then place the metal tip of the sonicator into the mixture

3. Ultra-sound waves of the sonicator stirred mixtures 4–5rpm (revolutions per min) for approx. 4 minutes.

4. Remove beaker from sonicator, filter using the Buchner funnel.

5. Place shaved ice into cooling condenser shaft of rotor-vapor.

6. Heat water in basin to approximately 70°-80°

7. Filtrate from sonicator was then vaporized until film is formed at the bottom of the round flask

8. Remove flask; add 10ml acetone saturated the film and then pipetted and transferred into a vial to be analyzed.

Florisil Cleanup

1. A piece of glass wool is placed into a column forming the base layer of a chromatography column attached to a ring stand.

2. Approximately 3g of sand is added

3. 2/3 of the column is filled with florisil to form the 3rd layer

4. Petroleum ether was drained into the column until saturated all 3 layers and liquid level reaches 1/2cm above the florisil

5. Soil equate collected from rota-vapor was poured into the chromatography column; the sample vial rinsed with petroleum ether to be poured into the column.

6. 100mls of 15% ethyl ether /75% petroleum ether was poured gradually into the column and allowed to drain.

7. 15% ethyl ether /75% petroleum ether was collected in a 125ml Erlenmeyer flask

8. 50% ethyl ether /50% petroleum ether was poured into the column and allowed to drain

9. Collect in another Erlenmeyer flask place aside (it has a polarity difference from the first solvent

10. The 15% ethyl ether / 75% petroleum ether, soil eluate was then transferred to a round bottom flask (rinsed with acetone and 15% ethyl ether / 75% petroleum ether) and placed into rota-vapor to be concentrated

11. The round bottom flask was then rinsed with about 5mls of 15% ethyl ether /75% petroleum ether and collected in sample vial

12. Repeat the above steps with the 50% ethyl ether/50% petroleum ether- soil elite sample. Using 50% ethyl ether either to rinse the round bottom flask as a schedule.
